چکیده
What is ELAV? The RNA-binding protein encoded by the elav gene of Drosophila. The name elav — pronounced ella-vee — is derived from initial mutant phenotypes: embryonic lethality and abnormal visual system. It is the founding member of a family of RNAbinding proteins. All members of the ELAV/Hu family have three ‘RNA recognition motif’ (RRM) domains, a hinge domain separating the first two RRMs from the third, and an aminoterminal domain (Figure 1A).

Members of the ELAV family of proteins contain three RNA recognition motifs (RRMs), which are highly conserved. ELAV, a Drosophila melanogaster member of this family, provides a vital function and exhibits a predominantly nuclear localization. To investigate if the RNA-binding property of each of the ELAV RRMs is required for ELAV's in vivo function, amino acid residues critical in RNA binding ...
متن کاملShared RNA-binding sites for interacting members of the Drosophila ELAV family of neuronal proteins
The product of the Drosophila embryonic lethal abnormal visual system is a conserved protein (ELAV) necessary for normal neuronal differentiation and maintenance. It possesses three RNA-binding domains and is involved in the regulation of RNA metabolism. The long elav 3'-untranslated region (3'-UTR) is necessary for autoregulation. We used RNA-binding assays and in vitro selection to identify t...
متن کاملNeural specificity of the RNA-binding protein Elav is achieved by post-transcriptional repression in non-neural tissues.
Drosophila Elav is the founding member of the conserved family of Hu RNA-binding proteins (RBPs), which play crucial and diverse roles in post-transcriptional regulation. Elav has long served as the canonical neuronal marker. Surprisingly, although Elav has a well-characterized neural cis-regulatory module, we find endogenous Elav is also ubiquitously transcribed and post-transcriptionally repr...
متن کاملELAV-mediated 3'-end processing of ewg transcripts is evolutionarily conserved despite sequence degeneration of the ELAV-binding site.
Regulation of alternative mRNA processing by ELAV (embryonic lethal abnormal visual system)/Hu proteins is mediated by binding to AU-rich elements of low complexity. Since such sequences diverge very rapidly during evolution, it has not been clear if ELAV regulation is maintained over extended phylogenetic distances. The transcription factor Erect wing (Ewg) is a major target of ELAV in Drosoph...
متن کاملELAV multimerizes on conserved AU4-6 motifs important for ewg splicing regulation.
ELAV is a gene-specific regulator of alternative pre-mRNA processing in Drosophila neurons. Since ELAV/Hu proteins preferentially bind to AU-rich regions that are generally abundant in introns and untranslated regions, it has not been clear how gene specificity is achieved. Here we used a combination of in vitro biochemical experiments together with phylogenetic comparisons and in vivo analysis...
متن کاملELAV, a Drosophila neuron-specific protein, mediates the generation of an alternatively spliced neural protein isoform
BACKGROUND Tissue-specific alternative pre-mRNA splicing is a widely used mechanism for gene regulation and the generation of different protein isoforms, but relatively little is known about the factors and mechanisms that mediate this process. Tissue-specific RNA-binding proteins could mediate alternative pre-mRNA splicing. In Drosophila melanogaster, the RNA-binding protein encoded by the ela...
